It's poorly worded but I think what they mean to say is, say for example your stock count in the inventory worksheet for a particular item is 10 - if it was set to First in Day, then the inventory transactions for that day are not "included" in the 10 so they will change the number. But if it you choose Last in Day, then that number 10 "includes" the transactions for that date, so they will not considered separately.
